Category:2000s Tamil-language filmsThis project is designed to study the mechanism(s) of action of retinoids in the regulation of connective tissue metabolism. The objectives are (1) to elucidate the mechanism of retinoid actions on collagen synthesis, (2) to define the role of transcriptional, translational, and posttranslational control of protein synthesis in retinoid modulation of connective tissue metabolism and (3) to determine the mechanism(s) involved in the nuclear translocation and inhibition of collagen biosynthesis by retinoids in cultured fibroblasts. These objectives will be accomplished by use of the following experimental strategies: (1) study of the synthesis of specific collagens by cultured fibroblasts in the presence of retinoids, (2) identification of mRNA's for type I, II and III procollagen and their retinoid modulation, (3) localization of retinoid receptors and induction of nuclear translocation of the receptor proteins, (4) effect of retinoids on posttranslational processing of procollagen, (5) effect of retinoids on the activity of RNA polymerase II and (6) characterization of specific RNA sequences which bind retinoid receptor proteins. Knowledge of these processes will provide an understanding of the mechanism(s) of action of retinoids in the regulation of connective tissue metabolism and may lead to the development of therapeutic agents for modulating connective tissue metabolism.Q:
